Path-clamp technique has been used to investigate potassium currents in
the membranes of single cardiomyocytes differentiated in vitro from
embrionic stem cells (D3 line). Changes in the expression of potassium
channels in early cardiomyogenesis have been shown. A significant role
of the channels of rapid transient current (Ito) has been proved in
forming and maintaining the stable electric activity of early
cardiomyocytes – from two to four days after the spntaneous contractions
begin. It has been found that unlike other types of potassium channels,
ATP-dependent channels are expressed practically in all cardiomyocytes
irrespective of the differentiation time, and that the density of IK,ATP
practically does not change at different stages. The role of
ATP-sensitive channels in variations that occur with the change of
energy metabolism of a cell has been shown. The mechanisms of the
influence of outside K+ concentration on the functioning of potassium
channels of the rapid transient current caused by ion-conformational
interaction of channel-forming protein with passing ions have been
determined.
