A.I. SEREBRYANIK. The ribosomal ATPase from higher eukaryotes.

– Manuscript

Thesis for a degree of Doctor of Philisophy (PhD in Biology, Special
Option – Molecular Biology.- Institute of Molecular Biology and
Genetics, National Academy of Sciences of Ukraine, Kyiv, 1998.

It has been first shown that highly purified ribosomes from higher
eukaryotes are strongly associated with ATPase activity, and its
properties have been studied. Two active sites have been revealed, one
of them may be stimulated by aminoacyl-tRNA. The data obtained show
participation of the ATPase in the regulation of the ribosomal A-site
responsible for the selection of cognate tRNA. A comparative analysis of
the ribosomal ATPase and yeast EF-3 has elucidated some common
properties. Higher eukaryotic ribosomes, even though possessing
intrinsic ATPase, are found to preserve in the course of evolution an
enhancing element for the yeast EF-3 ATPase stimulation. The thesis
contributes to a new understanding of detailed mechanisms of the
elongation cicle on 80S ribosome, including the elucidation of novel
points of protein biosynthesis regulation in mammals.
